Spender had strong friendships with some of the key figures in 20th century artistic life - including T. S. Eliot, W. H. Auden, W. B. Yeats, Christopher Isherwood, Cecil Day Lewis, Ernest Hemingway, Virginia and Leonard Woolf, George Orwell and former Poet Laureate Ted Hughes (an active founder member of the Stephen Spender Memorial Trust).

Spender was also friends with Isaiah Berlin, Igor Stravinsky, André Gide, Aldous Huxley, André Malraux, Louis MacNiece, Czeslaw Milosz, Octavio Paz, Joseph Brodsky, David Gascoyne, Bernard Spencer, Lytton Strachey, the Sitwells (Sacheverell, Sir Osbert, and Dr Edith), Edwin and Willa Muir, Pablo Neruda, Henry Moore, Frank Auerbach, Francis Bacon and David Hockney.
